These cell lines can be rendered non-metastatic (4T1-PE and 4T1.2-PE) by treatment with TARC-PE38 chemotoxin. RNA was extracted from both of these cell lines as well as the TARC-PE38 treated cell lines using Qiagen RNeasy kits. The total RNA from 3 replicates of each group was labeled using Illumina TotalPrep RNA Amplification Kits from Ambion. 0.75ug of this biotin-labeled cRNA was hybridized at 58 degrees C for 16 hours to Illumina's Sentrix MouseRef-8 v1.1 Expression BeadChips, washed, and the biotin labeled cRNA was detected by staining with streptavidin-Cy3.
