Title: Quantitative RT-PCR profiling of the nuclear receptor superfamily across fifty human cancer cell lines derived from nine different tissues

With the goal of defining subgroups of receptors that can be therapeutically targeted, we have profiled the expression of all 48 human NRs by quantitative RT-PCR in over fifty human cancer cell lines derived from nine different tissues. RNA was extracted using Rneasy (Qiagen). Genomic DNA contamination was eliminated by DNAase treatment. For each sample, RNA prepared from cell pellets received under an angreement with the NCI was used. Individual NR and 18S rRNA expression levels were measured in triplicate wells of a 384-well reaction plate with 10ng cDNA per well. The instrument was an Applied Biosystems 7900HT and the chemistry used was TaqMan. Data analysis was done using the efficiency-corrected ΔCT method. The normalizing gene was 18S rRNA.
